The Mogu Rune of Fate, when used, gives players a second shot at a chosen boss's loot table. When a raid group defeats a boss, each player in the raid will be prompted to use one of their Mogu Runes for a chance for extra loot. Internally, when players use a Mogu Rune, the game rolls a random number for each player, and if that number is above a certain threshold, it rewards them with a random spec-specific item off the boss's loot table. If the internal roll isn't high enough, the player receives just gold.

The Mogu Rune of Fate can only be used on tier 15 bosses as well as on the world boss Oondasta, and Nalak. In patch 5.4 Mogu Runes of Fate will have a cap of 20, and you'll be able to purchase a Mogu Rune of Fate for 1000 Timeless Coin each on the Timeless Isle. You can no longer get Runes from quests.
